【摘要】目的
比较高脂喂养的apoE基因敲除小鼠主动脉窦部和头臂动脉粥样硬化斑块的病理组织学状
况。方法选取6周龄雄性纯合子anpE基因敲除小鼠30只,均予以西方饮食喂养,分别在30周龄、42周龄、54周龄、66周龄、78周龄时,随机各取6只,取主动脉窦和头臂动脉做病理检测。酶法检测血脂情况,冰冻切片光镜下观察主动脉窦部和无名动脉粥样硬化斑块病理情况,vonkossa染色观察斑块钙化情况。结果
高脂喂养可使apoE基
因敲除小鼠迅速形成广泛且典型的动脉粥样硬化斑块,随时间进展,斑块体积逐渐增加,钙化逐渐加重;头臂动脉斑块出现较主动脉窦早且更加明显,斑块内包埋纤维帽,在主动脉窦斑块未见,但斑块钙化以主动脉窦部斑块最显著。结论高脂喂养apeE基因敲除小鼠是较为理想的动脉粥样硬化动物模型,头臂动脉是观察斑块不稳定的较为理想的窗口,而研究比较稳定斑块钙化,主动脉窦部更为理想。
