四种常见海参对小鼠肠道粘膜免疫功能
的保护作用

摘 要:目的:研究四种常见海参对环磷酰胺所致肠粘膜损伤的保护作用,并初步探讨其保护机制。方法:
70只BALB/C小鼠随机分为7地瓜组。采用腹腔注射环磷酰胺构建肠道粘膜损伤小鼠模型。检测胸腺、脾指数,
制备小肠切片并进行组织学观察,统计绒毛(villus)长度、隐窝(cryptV/C),酶
联免疫法(Elisa)检测肠粘膜SIgA、IFN-γ、IL-4的含量并统计γ)检
测小肠IgAγ/IL-4平衡。仿刺参与

Abstract:Purpose: To investigate the protective effect and mechanisms of four common species of sea cucumbers
on cyclophosphamide(CTX) induced intestinal mucosa damage. Methods: Seventy mice were randomly divided
into seven groups, including wild type group, model group, positive control group, Apostichopus japonicus group,
Isostichopus badionotus group, Thelenota ananas group and Acaudina molpadioidea group. Model of intestinal
mucosa damaged mice were induced by CTX injection. The change of the bodyweight, spleen index, thymus index,

villus length, crypt depth and V/C were detected. SIgA, IFN-γ and IL-4 content in intestinal mucosa were detected
by Elisa and IFN-γ/IL-4 was calculated. Protein expression of IgA were detected by Western Blot. Results:
Compared with the model group mice, spleen index, V/C and IFN-γ/IL-4 were all improved in the mice of all these
four sea cucumbers groups. The secretion of SIgA were dramatically enhanced in the mice of Apostichopus
japonicus and Thelenota ananas groups compared to the mice of model group, which was related with the
enhancement of IgA expression. Conclusion: Four common species of Sea Cucumbers can significantly protect the
small intestine mucosa of mice from the CTX induced damage, and the protective effects were more obvious in the
mice of Apostichopus japonicus and Thelenota ananas groups.

肠道不仅是机体的消化吸收器官,同时还是机体最大的“天然免疫器官”。肠道粘膜本身的结构入机体内部。小肠绒毛(villus)长度、
隐窝(crypt)深度及V/C比值可以反映小肠的
分泌型IgA(Secretory immunoglobulin A,SIgAdimeric IgA,dIgA)、综合功能状态[1]。
SC是pIgRJ链分子和分泌成分(secretory component,SC2(多聚免疫球蛋白受体)的细胞外段。SIgA此外,T细胞(helper T cell,发生[3]。
Th)对机体免疫具有重要的调节作用。其中-γ(和白细胞介素-2(IL-2)
等,Th2细胞因子包括白细胞介素-4(IL-4Th1/Th2系统平衡,从而
维持机体的正常免疫功能 [4]。

1 材料与方法
1.1 材料与试剂
1.1.1 海参
试验用海参原料为仿刺参(Apostichopus japonicus)、美国肉参(Isostichopus badionotus)、梅花参
(Thelenota ananas)和海地瓜(Acaudina molpadioidea),均为干品,购自青岛市南山水产市场。干海
参洗净、剪碎、烘干后粉碎机粉碎。海参粉过140目标准筛,细粉置于干燥器中保存。实验时加生理
盐水配成所需浓度的海参悬浊液。
1.1.2 实验动物
SPF级BALB/C小鼠,雄性,70只(6~8周龄,体重18g-22g),购自北京维通利华实验动物技术
有限公司,许可证号:SCXK(京)2007-0001;

贞芪扶正颗粒:甘肃扶正药业科技股份有限公司生产,批号:120601;环磷酰胺:江苏恒瑞医药
股份有限公司生产,批号:12042725;甲醛溶液,氯化钠,磷酸二氢钠,磷酸氢二钠,均为国产分析
纯;SDS,甘氨酸,Tris,丙烯酰胺,APS,北京索莱宝科技有限公司;医用X光胶片,Kodak;PVDF
膜(Millipore,美国);脱脂奶粉,(BD公司,美国);Mouse SIgA Elisa Kit(武汉USCNK,中国);
Mouse IL-4 Elisa Kit,Mouse IFN-γ Elisa Kit(R&D Systems,美国);抗体:mouse monoclonal IgG
anti-β-actin,goat anti- mouse IgA-HRP (Santa Cruz Biotechnology, 美国)。
